Короткий огляд (реферат): У дисертаційній роботі обґрунтовано схему профілактичних заходів за бактеріозів птиці, доведена доцільність використання екологічно-безпечного біоциду «ОЛЛДІЗ HPPA L» у системі дезінфекції птахівничих приміщень з метою підвищення біобезпеки, профілактики бактеріальних і вірусних інфекцій, а також покращення продуктивності птиці. Особлива увага приділялася комплексній оцінці мікробного фону в умовах промислового птахівництва; вивченню чутливості ізольованої мікрофлори до антибактеріальних препаратів, в тому числі до екстракту гінго білоба; визначенню параметрів токсичності, бактерицидної, фунгіцидної, віруліцидної, дезінвазійної активності дезінфекційного засобу «ОЛЛДІЗ HPPA L», обґрунтовано економічну ефективність використання засобу «ОЛЛДІЗ HPPA L » порівняно з вітчизняними та імпортними аналогами з урахуванням затрат на дезінфекцію, збереження продуктивності та біобезпеки поголів’я. Також акцентували увагу на вивченні властивостей вітамінного препарату «Аспір 35», як антистресу, та обґрунтуванні системи превенції бактеріальних хвороб птиці. Дослідження виконувались на базі Сумського національного аграрного університету, сертифікованого віварію Регіонального центру «ЕКОМЕДХІМ» СумДУ, наукової лабораторії науково-виробничої «Бровафарма» (м. Бровари, Київська обл.), Сумської регіональної державної лабораторії Держпродспоживслужби. Виробничі дослідження проведено в промислових птахогосподарствах різних форм власності Сумської області. Матеріалами дослідження були ізольовані мікроорганізми, патологічний матеріал, що відібраний від хворої та загиблої птиці, проби повітряного середовища птахівничих об’єктів, проби води та змиви з обладнання. Проведено мікробіологічний моніторинг виявлення збудників бактеріальних хвороб які циркулюють в птахівничих господарствах. У процесі дослідження встановлено, що найбільший відсоток виділених ізолятів становили представники роду Escherichia (30,1 %) та Salmonella spp. (14,9 %), що свідчить про домінування даних збудників у досліджених господарствах. Крім того були ізольовані мікроорганізми що належали до Klebsiella spp. (15,4 %), Enterobacter spp. (11,2 %), Yersinia spp. (10,5 %), Proteus spp. (7,1 %), Citrobacter spp. (6,2 %), Campylobacter spp. (2,5 %), Pseudomonas aeruginosa (1,2 %) і Clostridium spp. (0,9 %). Для ізоляції мікроорганізмів використовували загальноприйняті методи та серійні тест-системи Compact Dry (Total, EC, SL, Listeria, Yersinia), RIDA®STAMP (S. aureus, Staph, Pseudomonas). Встановлено асоціації мікроорганізмів, зв’язок між віком птиці, системою утримання та мікробним фоном. В результаті досліджень встановлено тенденцію до зростання рівня полірезистентності ізольованих мікроорганізмів до антибактеріальних препаратів, які раніше вважалися ефективними засобами терапії першого вибору. Виявлене зниження чутливості збудників одночасно до кількох груп антибіотиків суттєво обмежує можливості вибору ефективної етіотропної терапії. Отримані результати підкреслюють необхідність систематичного моніторингу антибіотикорезистентності, впровадження принципів раціонального застосування антимікробних засобів у ветеринарній практиці та розробки нових ефективних препаратів і засобів для проведення ветеринарно санітарних заходів. Дослідами доведено виражену антибактеріальну активність екстрактів Ginkgo biloba щодо штамів різних видів мікроорганізмів, зокрема Enterococcus faecalis, Proteus vulgaris, Serratia marcescens, Yersinia enterocolitica, Klebsiella pneumoniae, Pseudomonas aeruginosa, Campylobacter jejuni, Staphylococcus epidermidis, Staphylococcus aureus, Bacillus subtilis, Listeria innocua та Listeria monocytogenes. Отримані результати свідчать про перспективність використання екстрактів Ginkgo biloba як альтернативного превентивного засобу. Вперше були вивчена ефективність біоциду «ОЛЛДІЗ HPPA L» щодо збудників заразних хвороб, встановлено доцільність застосування його для профілактичної, поточної та вимушеної дезінфекції об’єктів ветеринарного нагляду в умовах птахівничих господарств. Дослідження також надало нові дані щодо токсикологічного профілю препарату зокрема, гострої та хронічної токсичності що дозволяє обґрунтовано рекомендувати його застосування у виробничих умовах. Визначено що дезінфікуючих засіб «ОЛЛДІЗ HPPA L» належить до IV класу токсичності згідно до Міжнародного стандарту ГОСТ 12.1.007-76, або до категорії 5 за Міжнародною глобальною класифікацією Global Harmonized System, (GHS). У ході дослідження місцевої подразнюючої дії препарату «ОЛЛДІЗ HPPA L» було встановлено, що аплікація 0,002 % розчину на шкіру кролів не викликала ознак запалення, гіперемії чи ушкодження шкірного покриву. Індекс сумарного подразнення в дослідній групі становив у середньому 0,31 ± 0,02 бала, що свідчить про відсутність клінічно вираженого подразнення та дозволяє за міжнародною класифікацією віднести його як безпечний (5 категорія) клас токсичності згідно GHS (міжнародною глобальною класифікацією). Наступним етапом дослідження було вивчення віруліцидної активності екологічно-безпечного дезінфікуючого засобу «ОЛЛДІЗ HPPA L» щодо збудників вірусних інфекцій, які мають значне епізоотичне значення у птахівництві. Віруліцидна дія проявляється як щодо РНК-вмісних, так і ДНК вмісних вірусів, з пролонгованим ефектом при оптимальних концентраціях (≥ 0,5 %). Вивчено антимікробну активність дезінфікуючого засобу «ОЛЛДІЗ HPPA L» у лабораторних умовах із використанням тест-культур бактеріальних збудників E. coli (штам ATCC 25922 F50) та S. aureus (штам ATCC 25923) у різних розведеннях, з експозицією 10 і 30 хв, за умов з білковим навантаженням та без нього. Встановлено, що фенольний коефіцієнт «ОЛЛДІЗ HPPA L» був значно вищим, ніж у контрольного зразка фенолу, і становив до 44,3 при короткочасній експозиції. Додавання білкових домішок (інактивована сироватка великої рогатої худоби) дещо знижувало ефективність препарату, про що свідчив білковий індекс на рівні 1,4-2,0. На наступному етапі експерименту досліджено ефективність препарату на твердих поверхнях (бетон, цегла, кахель, нержавіюча сталь), штучно контамінованих культурами E. coli та S. aureus. Обробка розчинами різної концентрації (0,01-0,1 %) упродовж 10-60 хв показала, що повна інактивація мікроорганізмів досягалася при концентрації 0,05 % вже за 30 хв, незалежно від типу поверхні. Дослідження ефективності екологічно-безпечного дезінфікуючого засобу «ОЛЛДІЗ HPPA L» у виробничих умовах проводили в умовах птахофабрики. Отримані результати свідчать, що «ОЛЛДІЗ НРРА L» має найбільш економічно обґрунтовані показники при збереженні ефективної дії, що підтверджує доцільність його впровадження у практику санітарної обробки птахівничих господарств. При вивченні антистресо-вітамінного препарату «Аспір-35» встановлено, що відповідно до вимог ГОСТ 12.1.007-76 він належить до 4 класу небезпеки (малонебезпечні речовини), а згідно з Глобальною гармонізованою системою класифікації та маркування хімічних речовин (GHS) – до 5 категорії токсичності. Доведено ефективність препарату Аспір 35 з метою превенції стресів в тому числі теплового. На основі матеріалів дисертаційного дослідження розроблено науково методичні рекомендації «Профілактика хвороб птиці за допомогою деззасобу «ОЛЛДІЗ НРРА L». Результати досліджень впроваджено в освітній процес факультету ветеринарної медицини Сумського національного аграрного університету та використовуються під час викладання дисциплін «Ветеринарна гігієна» та «Інфекційні хвороби птиці» для здобувачів другого (магістерського) рівня вищої освіти. За результатами проведених досліджень отримано патент України на корисну модель «Спосіб превенції заразних хвороб бройлерів», що підтверджує практичну цінність та інноваційний характер розроблених профілактичних заходів.
Опис: The dissertation work substantiates the scheme of preventive measures for poultry bacteriosis, proves the feasibility of using the environmentally safe biocide "ALLDIZ HPPA L" in the disinfection system of poultry premises in order to increase biosafety, prevent bacterial and viral infections, as well as improve poultry productivity. Special attention was paid to a comprehensive assessment of the microbial background in industrial poultry farming; study of the sensitivity of isolated microflora to antibacterial drugs, including ginkgo biloba extract; determination of toxicity parameters, bactericidal, fungicidal, virulicidal, disinvasive activity of the disinfectant "ALLDIZ HPPA L", substantiates the economic efficiency of using the product "ALLDIZ HPPA L" compared to domestic and imported analogues, taking into account the costs of disinfection, maintaining productivity and biosecurity of livestock. Attention was also focused on studying the properties of the vitamin preparation "Aspire 35" as an anti-stress agent, and substantiating the system for preventing bacterial diseases in poultry. The research was carried out on the basis of Sumy National Agrarian University, the certified vivarium of the Regional Center "EKOMEDHIM" of Sumy State University, the scientific laboratory of the scientific and production "Brovapharma" (Brovary, Kyiv region), the Sumy Regional State Laboratory of the State Service for Food Safety and Consumer Protection. Production research was conducted in industrial poultry farms of various forms of ownership in Sumy region. The research materials were isolated microorganisms, pathological material taken from sick and dead birds, samples of the air environment of poultry facilities, water samples and washdowns from equipment. Microbiological monitoring was carried out to detect pathogens of bacterial diseases circulating in poultry farms. During the study, it was found that the largest percentage of isolated isolates were representatives of the genus Escherichia (30.1%) and Salmonella spp. (14.9%), which indicates the dominance of these pathogens in the studied farms. In addition, microorganisms belonging to Klebsiella spp. (15.4%), Enterobacter spp. (11.2%), Yersinia spp. (10.5%), Proteus spp. (7.1%), Citrobacter spp. (6.2%), Campylobacter spp. (2.5%), P. aeruginosa (1.2%) and Clostridium spp. (0.9%) were isolated. For the isolation of microorganisms, generally accepted methods and serial test systems Compact Dry (Total, EC, SL, Listeria, Yersinia), RIDA®STAMP (S. aureus, Staph, Pseudomonas) were used. Associations of microorganisms, the relationship between the age of the bird, the housing system and the microbial background were established. As a result of the research, a trend towards an increase in the level of polyresistance of isolated microorganisms to antibacterial drugs, which were previously considered effective first-choice therapy, has been established. The revealed decrease in the sensitivity of pathogens simultaneously to several groups of antibiotics significantly limits the possibilities of choosing effective etiotropic therapy. The results obtained emphasize the need for systematic monitoring of antibiotic resistance, the implementation of the principles of rational use of antimicrobial agents in veterinary practice, and the development of new effective drugs and means for carrying out veterinary and sanitary measures. The experiments proved the pronounced antibacterial activity of Ginkgo biloba extracts against strains of various types of microorganisms, in particular Enterococcus faecalis, Proteus vulgaris, Serratia marcescens, Yersinia enterocolitica, Klebsiella pneumoniae, Pseudomonas aeruginosa, Campylobacter jejuni, Staphylococcus epidermidis, Staphylococcus aureus, Bacillus subtilis, Listeria innocua and Listeria monocytogenes. The obtained results indicate the prospects of using Ginkgo biloba extracts as an alternative preventive agent. For the first time, the effectiveness of the biocide "ALLDIZ HPPA L" against pathogens of infectious diseases was studied, the feasibility of its use for preventive, current and forced disinfection of veterinary supervision facilities in poultry farms was established. The study also provided new data on the toxicological profile of the drug, in particular, acute and chronic toxicity, which allows us to reasonably recommend its use in production conditions. It was determined that the disinfectant "ALLDIZ HPPA L" belongs to the IV toxicity class according to the International Standard GOST 12.1.007-76, or to category 5 according to the International Global Harmonized System (GHS). During the study of the local irritant effect of the drug "ALLDIZ HPPA L" it was found that the application of a 0.002% solution to the skin of rabbits did not cause signs of inflammation, hyperemia or damage to the skin. The total irritation index in the experimental group was on average 0.31±0.02 points, which indicates the absence of clinically pronounced irritation and allows it to be classified as safe (category 5) of toxicity according to the GHS (international global classification). The next stage of the study was the study of the virucidal activity of the environmentally safe disinfectant "ALLDIZ HPPA L" against pathogens of viral infections that have significant epizootic significance in poultry farming. The virucidal effect is manifested both against RNA-containing and DNA-containing viruses, with a prolonged effect at optimal concentrations (≥0.5%). The antimicrobial activity of the disinfectant "ALLDIZ HPPA L" was studied in laboratory conditions using test cultures of bacterial pathogens E. coli (strain ATCC 25922 F50) and S. aureus (strain ATCC 25923) in various dilutions, with exposure of 10 and 30 min, under conditions with and without protein loading. It was found that the phenol coefficient of "ALLDIZ HPPA L" was significantly higher than that of the control sample of phenol, and was up to 44.3 with short-term exposure. The addition of protein impurities (inactivated bovine serum) slightly reduced the effectiveness of the drug, as evidenced by the protein index at the level of 1.4-2.0. At the next stage of the experiment, the effectiveness of the drug on hard surfaces (concrete, brick, tile, stainless steel) artificially contaminated with E. coli and S. aureus cultures was studied. Treatment with solutions of various concentrations (0.01-0.1%) for 10-60 min showed that complete inactivation of microorganisms was achieved at a concentration of 0.05% in 30 min, regardless of the type of surface. Research into the effectiveness of the environmentally safe disinfectant "ALLDIZ HPPA L" in production conditions was carried out in a poultry farm. The results obtained indicate that "ALLDIS NRPA L" has the most economically justified indicators while maintaining effective action, which confirms the feasibility of its introduction into the practice of sanitary treatment of poultry farms. When studying the anti-stress vitamin preparation "Aspire-35", it was found that, in accordance with the requirements of GOST 12.1.007-76, it belongs to the 4th hazard class (low-hazard substances), and according to the Globally Harmonized System of Classification and Labeling of Chemicals (GHS) - to the 5th toxicity category. The effectiveness of the preparation Aspir 35 for the prevention of stress, including heat stress, has been proven. Based on the materials of the dissertation research, scientific and methodological recommendations "Prevention of poultry diseases using the disinfectant "ALLDIS NRPA L" have been developed. The results of the research were implemented in the educational process of the Faculty of Veterinary Medicine of Sumy National Agrarian University and are used when teaching the disciplines "Veterinary Hygiene" and "Infectious Diseases of Poultry" for applicants of the second (master's) level of higher education. Based on the results of the research, a patent of Ukraine was obtained for a useful model "Method for the prevention of infectious diseases of broilers", which confirms the practical value and innovative nature of the developed preventive measures.
